Kanglaite enhances nedaplatin-induced inhibition of cell viability, migration, and epithelial-mesenchymal transition in cervical cancer cells

Abstract
OBJECTIVE: To investigate whether Kanglaite (KLT), a main ingredient of Coixseed oil (a traditional Chinese medicine), enhances nedaplatin (NDP)-induced antitumor activity of cervical cancer. STUDY DESIGN: C-33A and Hela cells were treated with NDP and/or KLT in vitro. Cell viability was determined by CCK-8 assay. Cell migration was measured by wound healing assay. The expression levels of epithelial-mesenchymal transition (EMT)-related protein E-cadherin and N-cadherin were detected by western blot. RESULTS: Our results revealed that KLT dramatically inhibited cell viability of cervical cancer cells in a doseand time-dependent manner. In addition, we found that KLT or NDP treatment notably reduced cell viability and inhibited cell migration and EMT of cervical cancer cells. Furthermore, the administration of KLT prior to or at the same time as NDP treatment inhibited cell cytotoxicity, cell migration, and EMT of cervical cancer cells as compared with the single NDP treatment. CONCLUSION: These results demonstrate that KLT enhanced NDP-induced inhibition of cell viability, migration, and EMT in cervical cancer cells. These results indicate that KLT could be used as an adjuvant treatment for cervical cancer chemotherapy. © 2018 Science Printers and Publishers, Inc.
